Nandamuri is the son of former actor and Chief Minister of Andhra Pradesh, Nandamuri Taraka Rama Rao.
He began his acting career as a child artist in 1972 in the Telugu film Pistolwali.
In 1987, he had a cameo role in the Telugu film Trimurtulu, directed by K. Murali Mohan Rao.
He is an active member of the Telugu Desam Party founded by his father.
He holds the record for appearing in the highest number of double roles in the Telugu film Adhinayakudu (2012).

He serves as the chairman of the trust board of the Basavatarakam Indo American Cancer Hospital & Research Institute in Hyderabad, India.
